Antifreeze Evaluation of Two Dehydrin Proteins from Pseudotsuga menziesii and Larix principis-rupprechtii Mayr

Dehydrins exist widely in plants and play an important role abiotic stress resistance. Two low-temperature-induced dehydrin-like genes, PmCAP LpCAP, from the pine species Pseudotsuga menziesii Larix principis-rupprechtii Mayr were cloned found to contain 576 bp 687 bp, encoding 191 228 amino acids, respectively. Both genes individually assembled into prokaryotic expression vectors transferred E. coli cells. When transgenic stains cultured at −5 °C, lethal time 50% (LT50) was 50 h 54 for respectively, compared with 32 control. −20 LT50 38 h, 41 25 PmCAP, Thermal hysteresis (TH) testing of LpCAP proteins revealed TH values 0.27 °C 0.72 relative 0.05 BSA These results indicate that two dehydrin have antifreeze characteristics their levels well relation environmental conditions growth (Larix mostly grows cold high-altitude zones, while temperate low-altitude zones). especially, could be a better gene resource molecular breeding plant
